【Animal modeling】-Effect of Lycium barbarum polysaccharide on tumor growth and metastasis in spontaneous breast cancer MMTV-PyMT mice

  Objective: To investigate the effect of Lycium barbarum polysaccharide on the growth and metastasis of breast cancer using MMTV-PyMT mice as a breast cancer model.

  Methods: Expansion group identified transgenic spontaneous breast cancer MMTV-PyMT mice, and positive female mice were randomly divided into LBP group and control group, with 8 mice in each group. /kg, intraperitoneal injection) treatment, the control group was given normal saline, and the tumor volume was measured once every 2 days. After 4 weeks, they were killed by dislocation. The number of metastatic nodules on the lung surface was observed after the lung was fixed with Bouin's solution. After the tumor tissue was fixed and embedded The proliferation of tumor cells and the density of blood vessels were detected by immunohistochemistry.

  Results: The tumor formation rate of MMTV-PyMT-positive mice was 100%. Compared with the control group, the tumor weight (4.208±0.4463 g) in the LBP group was significantly lighter than that in the control group (6.477±0.3724 g), and the tumor volume was also significantly smaller than that in the control group. The group was small; the number of lung surface nodules in the LBP group (12±1.155) was significantly less than that in the control group (20±2.745), and the results of immunohistochemical staining showed that the number of cell proliferation and microvessel density in the tumor tissue of the LBP group were significantly less than those of the control group .

  Conclusion: Lycium barbarum polysaccharide can inhibit the growth and metastasis of breast cancer in MMTV-PyMT mice, and inhibit tumor cell proliferation and angiogenesis. MMTV-PyMT mice can be used as a model of breast cancer lung metastasis for anti-tumor drug research.
