Objective: To establish a rat model of premature ovarian failure induced by Tripterygium wilfordii polyglycosides, and to study the effect of Bushen Yangxue Recipe on anti-mullerian in rats with premature ovarian failure.
Methods: Tripterygium wilfordii polyside was used to establish a rat model of premature ovarian failure. After successful modeling, high-dose, medium-dose, and low-dose Bushen Yangxue Recipe were administered to the rats by gavage, once a day, for a total of 30 d. After the administration, the morphology of the ovaries of the rats in each group was observed under a microscope. Enzyme-linked immunosorbent assay, immunohistochemical staining and quantitative reverse transcription polymerase chain reaction (qRT-PCR) were used to detect AMH content in serum and AMH protein and mRNA expression levels in ovaries, respectively.
Results: Histomorphology showed that the growth of follicles and corpus luteum in the high, middle and low dose groups were better than those in the model group. This results in a decrease in AMH concentration in rat serum and AMH expression level in ovary.
Conclusion: Bushen Yangxue Recipe can reverse the decrease of AMH level in ovary caused by Tripterygium wilfordii polyglycosides, suggesting that Bushen Yangxue Recipe has the effect of preventing premature ovarian failure.
