【Animal modeling】-Effect of Jiuxieling granules on the expression of TLR4 and NF-κB p65 in rats with ulcerative colitis of spleen and kidney yang deficiency

  Objective: To investigate the effect of Jiuxieling Granules on the expression of TLR4 and NF-κB p65 protein genes in colon tissue of rats with ulcerative colitis due to spleen and kidney yang deficiency.

  Methods: An animal model was established by oral administration of rhubarb decoction, intramuscular injection of hydrocortisone, combined with TNBS (2,4,6-trinitrobenzenesulfonic acid) mixed with ethanol for enema. Ninety rats were randomly divided into blank group , model group, Jiuxieling granule treatment group for 7, 14, 21 d and positive control (SASP) group, immunohistochemical SP method and RT-qPCR method were used to detect TLR4 and NF-κB in colon tissue of rats after treatment.   Results: The results of colon mucosa morphology score showed that the scores of each treatment group were decreased, and the difference was significant (P<0.01). significantly enhanced (P<0.01); after drug intervention, the expressions of TLR4 and NF-κB p65 in each treatment group were attenuated (P<0.05); RT-qPCR results showed that compared with the blank group, TLR4, NF-κB and NF-κB in the model group The expression of -κB p65 was enhanced (P<0.01); compared with the model group, the expressions of TLR4 and NF-κB p65 in each group were decreased after treatment (P<0.05).

  Conclusion: Jiuxieling granules can reduce the inflammatory response of colonic mucosa and play a role in repairing the mucosa. The reason is related to the decrease of TLR4 and NF-κB p65 gene transcription and protein expression in colon tissue of UC model rats with spleen and kidney yang deficiency.
