【Animal Modeling】-Effect of Qiangzhi Prescription on Rats with Multiple Tic Disorder and Fear Behavior

  OBJECTIVE: To explore the mechanism of Qiangzhi prescription in the treatment of multiple tic disorder (TS) with complex fear behavior.

  Methods: After intraperitoneal injection of iminodipropionitrile (IDPN) to establish a TS rat model, further acoustic and electrical stimulation was used to establish a TS compound fear rat model. The content of dopamine (DA) in brain tissue was detected by high performance liquid phase method (HPLC), the content of tyrosine hydroxylase (TH) was detected by immunohistochemistry, and the expression of TH mRNA was detected by RT-PCR.

  Results: Compared with the normal control group, the stereotyped behavior and motor behavior of the model control group increased, the freezing time was prolonged, and the DA, TH and TH mRNA contents in the brain tissue had no significant changes; The behavior and motor behavior decreased, the freezing time was shortened, and the DA, TH and TH mRNA contents in the brain tissue decreased.

  Conclusion: Qiangzhi group prescription can improve the tic-complex fear behavior in model rats, and its mechanism of action may be achieved by down-regulating the expression of TH mRNA, reducing the content of TH, and reducing the synthesis of DA.
