【Animal modeling】-Immune protection of Sap2 protein and dendritic cells against systemic Candida albicans infection

  Objective: Mice were immunized with recombinant protein and dendritic cells (DC) to observe the immune protection effect on systemic Candida albicans.

  Methods: Prokaryotic expression and induced expression of Candida albicans Sap2 protein; after sensitizing mouse DCs with Candida albicans spores and hyphae suspensions, mice in 5 groups were immunized three times with recombinant protein and sensitized DCs, sensitized DCs, DCs, Sap2, PBS, reconstructed systemic Candida albicans infection model, and observed the survival time of mice, kidney pathological sections, negative bacteria in the kidneys, and the percentage of CD4+ and CD8+ to observe the protection.

  RESULTS: The pET32a-Sap2 prokaryotic expression vector was successfully constructed, and the target protein was induced to express and collected and purified. Recombinant protein and sensitized DC, DC and Sap2 protein can effectively prolong the survival period of mice infected with systemic Candida albicans, protect the kidneys of mice, and enhance the immunity of mice, among which the multi-component combined immunization effect is better.

  Conclusion: Multi-component combined immunization can well resist systemic Candida albicans infection, which lays the foundation for the study of new multi-component combined vaccine.
