【Animal modeling】-The protective effect of Jiedu Quyu Zishen Decoction on the kidneys of MRL/lpr lupus mice by intervening TLR9 signaling pathway

Objective: Based on the intervention effect on TLR9-MyD88-NF-κB pathway, to clarify the protective effect of Jiedu Quyu Zishen Decoction on the kidneys of lupus mice.

Methods: Thirty MRL/lpr lupus mice were randomly divided into three groups: model control group, prednisone treatment group, and traditional Chinese medicine group. Each treatment group was given corresponding drug treatment for 8 weeks. HE staining, PAS staining and Masson staining were used to stain the kidney sections of MRL/lpr mice, respectively, to observe and compare the pathological changes and therapeutic effects of the three groups of mice; real-time PCR was used to observe and compare the TLR of each group of mice. -9. The difference of MyD88 and NF-κB mRNA expression.

RESULTS: Compared with the model group, the traditional Chinese medicine group could significantly improve the renal tissue pathological damage in MRL/lpr mice. The results of Real-time PCR showed that the mRNA expressions of TLR-9, MyD88 and NF-κB in the kidney tissue of MRL/lpr mice in the traditional Chinese medicine group were lower than those in the model group (all P<0.05).

Conclusion: Jiedu Quyu Zishen Decoction has a protective effect on the kidneys of MRL/lpr mice, and can affect its pathological changes, reduce kidney damage, and delay the course of the disease. -9-MyD88-NF-κB signaling pathway related inhibition.
