Objective To investigate the effect of β-arrestin2 gene knockout on the function of mouse peritoneal macrophages.
Methods The pMφ of wild type (wild type, WT) and β-arrestin2 knockout ( β-arrestin2- / - ) mice were isolated, and the effect of β-arrestin2 knockout on the migration of pMφ was detected by Transwell assay; The changes of phagocytic function of pMφ without β-arrestin2 gene were detected by experiments; the changes of CD86 on the surface of pMφ were detected by flow cytometry; the effects of β-arrestin2 gene knockout on the production of inflammatory factors in pMφ were detected by ELISA; Western Blot was used to detect β -expression of proteins related to the arrestin2, JAK1/STAT1 pathway.
Results Compared with the WT group, knockout of β-arrestin2 significantly reduced the migration ability of pMφ, enhanced the phagocytic function of pMφ, and up-regulated the expression of CD86 molecule on the surface of pMφ. At the same time, pMφ secreted inflammatory factors IL-1β, TNF-α, The level of IL-6 was also significantly increased; Western Blot results showed that the expressions of p-JAK1 and p-STAT1 were significantly up-regulated in pMφ lacking β-arrestin2.
Conclusion β-arrestin2 has regulatory effects on the migration, phagocytosis, polarization and other functions of mouse pMφ, and its mechanism may be related to the regulation of JAK1/STAT1 pathway.
