【Animal modeling】-Analysis of genetic quality of Wistar rat microsatellite DNA population based on group standard T/CALAS 21—2017

  Objective To analyze the genetic quality of Wistar rats in closed groups at different periods by using the method recommended by the group standard T/CALAS 21-2017, and to evaluate the applicability of the group standard T/CALAS 21-2017.

  Methods The Wistar rats selected in 2015 and 2019 were named as group A and group B, respectively. According to the requirements of the group standard T/CALAS 21-2017, 25 pairs of microsatellite primers were used to obtain the genetic parameters of the two groups of Wistar rats for quality analysis, and the polymorphism information content (PIC) was used to analyze the locus polymorphism. sex.

  Results 100 and 69 alleles were detected in rats in group A and group B, respectively. The mean heterozygosity of the two populations was 0.574 and 0.447, respectively, and the mean PIC was 0.541 and 0.393, respectively.

  Conclusion The group standard T/CALAS 21-2017 has good applicability in the genetic quality analysis of closed group rats, and the genetic diversity of Wistar rats in group A is better than that in group B.
