Objective To establish a non-invasive tracheal instillation method for rats with visual, simple, rapid and precise instillation.
Methods A 1 mL medical syringe and capillary were used to make a sampler, an optical fiber was used to realize visual operation, soapy water was used to test whether the tracheal intubation was correct, and the sampler was inserted into the trachea through a breathing tube to complete the instillation. Twenty rats were randomly divided into two groups and were instilled with 0.9% NaCl solution and fine particulate matter (PM2.5) suspension, respectively.
Results A total of 120 tracheal intubation and instillation operations were completed in 20 rats. 24 hours after the last instillation, the rat lung tissue was taken for HE staining. It was found that the lung tissue of the PM2.5 group showed obvious pathological changes, basically consistent with chronic obstructive disease. Characteristics of chronic obstructive pulmonary disease (COPD).
Conclusion The improved non-invasive tracheal instillation method in rats is simple, fast and safe, and does not require special equipment, and can be widely used in researches related to drug administration and exposure to the respiratory tract of rats.
