【Animal Modeling】Effect of -60Coγ-ray radiation on the spleen of naked mole rats

  Objective To observe the changes of spleen immune system and tissue biochemical indexes in naked mole-rats after 60Co γ-ray irradiation, and to preliminarily understand the radiation tolerance characteristics of naked mole-rats spleen.

  Methods Naked mole rats were irradiated with 60Co γ-rays with a dose rate of 1.163 Gy/min and a total dose of 10 Gy. The spleens of normal control and naked mole rats were collected at 7 d, 14 d and 21 d after irradiation, and the proportion of B lymphocytes and macrophages in the spleen was detected by flow cytometry. Animal tissue biochemical detection kits were used to detect malondialdehyde (MDA), total antioxidant capacity (T-AOC), glutathione (GSH) and oxidized glutathione (GSSG) in spleen tissue.

  Results The results of flow cytometry showed that the proportion of B lymphocytes and macrophages increased significantly after irradiation (P<001). Biochemical test results showed that the level of MDA in the spleen increased significantly after irradiation (P<0.05). GSH and GSSH gradually and significantly decreased (P < 0.05); T-AOC levels decreased significantly after irradiation (P < 0.01), but increased at 21 d after irradiation, which was significantly higher than at 7 d and 14 d after irradiation (both P < 0.01). ).

  Conclusion The spleen of naked mole rat has a certain degree of tolerance to 60Co γ-rays, but the tolerance mechanism is not yet clear.
