【Animal modeling】-Distribution of adenovirus type 5 in C57BL/6J mice after infection by different routes

  Objective To detect the distribution of adenovirus type 5 (Ad5) in C57BL6J after infection by four ways.

  Methods 112 C57BL/6J mice were divided into 4 groups. Each mouse was given 100 μL of 1×109 pfu/mL Ad5 solution by intranasal, gavage, tail vein and intraperitoneal injection. , 4d, 5d, 6d and 7d, 2 male and 2 mice in each group were sacrificed, and the lymph nodes, testis, epididymis, uterus, ovary, spleen, kidney, liver, lung, heart, brain, blood and bone marrow were collected for fluorescence quantitative PCR detection .

  Results Ad5 was detected in the lung and spinal cord tissues in the nasal drip group, and Ad5 in the lung tissue decreased significantly at 4 d after infection; Ad5 was not detected in all tissues in the gavage group; Ad5 was detected in the liver and spleen in the tail vein injection group Ad5, and the descending speed was relatively slow; Ad5 was detected in the spinal cord, spleen, epididymis and testis in the intraperitoneal injection group. The content of Ad5 in the spinal cord of the intraperitoneal injection group was about 10 times higher than that of the intranasal group, the content of Ad5 in the spleen was comparable to that in the spleen of the tail vein injection group, and the content of Ad5 in the epididymis and testis was only 1% of that detected in other positive tissues. ~10%.

  Conclusion Ad5 can be instilled intranasally every 3 days for lung infection test; tail vein injection can be used for liver infection test; tail vein injection or intraperitoneal injection method can be used for spleen infection test; and intraperitoneal injection method can be used for spinal cord infection test.
