Objective To detect the correlation between the SNP locus of SLCIA2 gene and the activity of Labrador retrievers and golden retrievers, in order to screen effective genetic molecular markers for the early selection and breeding of guide dogs.
Methods Ninety-nine dogs (80 Labrador retrievers and 19 golden retrievers) from the Guide Dog Training Base in Dalian, China were selected, and the passive test (PT) behavioral test method was used to evaluate the large activity. At the same time, the venous blood of the 99 dogs was collected, and the genomic DNA samples in the blood cells were extracted. The PCR/sequencing method was used to discuss the 99 dogs' SLCIA2 gene T129C, T1549G and T47IC 3 SNP loci for genotyping, and statistical analysis of the dogs' Correlation of activity with SLCIA2 SNP loci.
RESULTS: Among the three SNP loci of SLCIA2 gene, T129C and T1549G had no polymorphism, and only C/C type and G/G type existed, respectively. Three genotypes exist at the T47IC locus: T/T, T/C and C/C. There was no significant correlation between the three genotypes and activity, sex and breed (P>0.05). In addition, there was no significant correlation between breed and activity (P>0.05), but there was a significant correlation between gender and activity (P=0.049).
Conclusion The T129C, TI549G and T471C loci of SLCIA2 gene have no significant correlation with the large activity of guidance and can not be used as an index for genetic screening of guide dogs. Because there is a significant correlation between sex and activity, activity in dogs should be graded differently for males and females in behavioral testing.
