Objective To perform genetic testing on different production and breeding groups of cynomolgus monkeys in Guangxi, analyze the genetic background of cynomolgus monkeys in different breeding groups in Guangxi, and provide basic information for the establishment of genetic quality detection methods and population resource bank of cynomolgus monkeys in Guangxi.
Methods Twenty microsatellite loci and capillary electrophoresis were used to detect cynomolgus monkeys in three different production groups in Guangxi, and the genetic variation parameters within and between groups were calculated.
Results A total of 237 alleles were detected, and the observed number of alleles (Na) was 5-19, with an average of 11.85; the average expected heterozygosity (He) was 0.85; and the average polymorphic information content (PIC) was 0.817. Three different production groups (F, N and W) detected 158, 158 and 173 alleles, respectively, and the average expected heterozygosity (He) was 0.8371, 0.8318 and 0.8642, respectively; the average polymorphic information content (PIC) was 0.7692, 0.7653 and 0.8001, three different production groups had higher genetic polymorphisms. The majority of loci were in H-W equilibrium by Handy-Weinberg equilibrium test of the three production groups. The mean value of the inbreeding coefficient Fis within the group is 0.0235, the mean value of the inbreeding coefficient Fit of the total population is 0.0628, the mean value of the genetic differentiation coefficient Fs is 0.0402, and the mean value of the gene flow Nm is 5.9616, indicating that the three production groups are basically in the state of random mating, and the genetic differentiation between groups is very small. The genetic distance of the three production groups was 0.2556-0.3223, and the genetic similarity was 0.7245-0.7745. Cluster analysis showed that the F group and the N group were first clustered into one group, and then grouped with the W group, which was consistent with the three breeding monkey farms. Conclusion This study effectively analyzed the genetic polymorphisms and genetic relationships among the populations of Guangxi cynomolgus monkeys. The selected 20 microsatellite loci can be used for the genetic quality detection of cynomolgus monkeys.
