【Animal Modeling】-Programmed freezing of hatched blastocysts and their dormant embryos in mice-Differential expression of Mgst1 gene before and after thawing

  Objective To analyze the role of Mgst1 gene in programmed freezing of mouse embryos by comparing the relative expression of Mgst1 gene and the differential expression distribution of Mgst1 protein before and after programmed freezing and thawing of hatched blastocysts and their dormant embryos. , providing a new theoretical basis for the study of embryonic antifreeze mechanism.

  Methods Immunofluorescence images of the expression distribution of Mgst1 protein in embryos were collected by confocal laser scanning microscope; qRT-PCR technology was used to identify the relative expression of Mgst1 gene in embryos.

  Results The immunofluorescence images showed that Mgst1 protein was expressed in both hatched blastocysts and dormant embryos before and after freezing; qRT-PCR showed that there was no significant difference in the relative expression of Mgst1 gene between hatched blastocysts and dormant embryos before freezing ( P > 0. 05); the relative expression of Mgst1 gene in hatched blastocysts after freezing was significantly up-regulated (P < 0. 05); the relative expression of Mgst1 gene in dormant embryos after freezing was significantly up-regulated (P < 0.05). 0. 01); the relative expression of Mgst1 gene in frozen dormant embryos was significantly up-regulated compared with frozen hatched blastocysts (P < 0.05).

  Conclusion Mgst1 gene may play an important positive regulatory role in the antifreeze mechanism of mouse embryos.
