Objective To analyze the expression of IL-18 in the intestinal tissue of tree shrew and the changes of intestinal flora under the action of D-galactose.
Methods Ten male tree shrews were randomly divided into normal group and D-galactose group (n=5). The treatment group was injected with D-galactose (0.6 g/kg) by intraperitoneal injection for 8 weeks, and the normal group was injected with normal saline according to the same operation. The ileum, cecum and colon were isolated and their contents were collected, the expression of IL-18 was detected by immunohistochemistry, and the bacterial DNA sequence of intestinal contents was analyzed by 16S rDNA high-throughput sequencing.
Results The expressions of IL-18 in the ileum, cecum and colon of the D-galactose group were significantly increased (P<0.05). The abundance of Bifidobacterium in the colon decreased significantly.
Conclusion D-galactose increases the expression of intestinal inflammatory factor IL-18 and intestinal flora disturbance. It is speculated that the intestinal flora imbalance of tree shrew causes the increase of I-18 expression and promotes the intestinal inflammatory response.
