【Animal Modeling】-Evaluation of the detection method of intestinal mucosal barrier damage in diabetic rats and discussion of the hypoglycemic effect of propolis

  Objective: To analyze and evaluate the detection method of intestinal mucosal barrier function in diabetic rats, and to explore whether propolis exerts hypoglycemic effect by repairing intestinal mucosal barrier.

  METHODS: A diabetic rat model was established. Intervention with 80 and 160 mg/kg propolis continued for 4 weeks. Roche blood glucose meter was used to detect tail blood glucose (FBG), high performance liquid chromatography to detect erythrocyte glycosylated hemoglobin (HbA1c) content, and ELISA to detect plasma D-lactic acid. ( D-LA ) and zonulin ( zonulin ) contents were observed by transmission electron microscope , the ultrastructural changes of the junction apparatus between intestinal epithelial cells were observed , and the expression levels of tight junction related proteins were detected by Western Blotting

  RESULTS: Diabetic rat model was successfully established. After 4 weeks of intervention with 80 and 160 mg/kg propolis, the blood glucose levels of the rats reached 17.57 mmol/L and 18.24 mmol/L, and the HbA1c content reached 8.94% and 8.22%, respectively, which were higher than the model. The phenomenon of tight junctions between epithelial cells in the ileum and colon tissue of diabetic rats and abnormal widening of adhesive junction gaps in the ileum and colon tissue of diabetic rats after supplementation with propolis was effectively alleviated in the group (P<0 05). The expression level of ZO-1 protein was significantly increased in a dose-dependent manner ( P<0 05). Serum D-LA levels were 160 03 ng / mL and 151 18 ng / mL zonulin levels after 80 and 160 mg / kg propolis intervention, respectively. 650 14 μg/

  Conclusion: The observation of the ultrastructure of the intestinal mucosal barrier and the detection of the expression level of the tight junction-related proteins in the intestinal tissue may have higher sensitivity than the serological detection of the intestinal mucosal barrier function, which can be used as a reliable experimental method for the repair of the intestinal mucosal barrier in the hypoglycemic effect of propolis.
