OBJECTIVE: To verify whether the microsatellite marker detection method in the group standard is applied to the genetic quality detection of experimental animals, and whether it has good repeatability in three laboratories.
Methods: Three laboratories simultaneously used 12 microsatellite loci for PCR amplification of 6 experimental mouse DNAs, and the amplified products were subjected to agarose electrophoresis and sequencing.
Results: Beijing laboratory obtained clear electrophoresis bands and obtained 72 alleles; Zhejiang laboratory obtained clear electrophoresis bands and obtained 72 alleles; Guangdong laboratory obtained clear electrophoresis bands and obtained alleles 72. The sequencing results showed that 3 laboratories obtained alleles with the same fragment size.
Conclusion: The microsatellite marker detection method can realize the evaluation of the genetic quality of experimental animals, and obtain good repeatability in different laboratories.
