【Animal modeling】-PCR detection and analysis of various bacteria in experimental rats and mice

  Objective: To investigate the bacterial carriage status of rats and mice in Shanghai laboratory animal production facilities.

  Methods: Comparing domestic and foreign experimental animal bacterial monitoring programs, Helicobacter, Citrobacter rodentium, Corynebacterium bovis, Staphylococcus aureus, Pseudomonas aeruginosa and Klebsiella pneumoniae were selected as testing items, which were collected in 2014 and 2016, respectively A total of 848 samples were used to detect the above 6 pathogens by PCR.

  Results: There was no Staphylococcus aureus contamination in the SPF facilities, and no Klebsiella pneumoniae contamination in the clean facilities in 2014. Other pathogens were contaminated to varying degrees in different facilities. Helicobacter and bovine rod were found in all clean facilities. Bacillus contamination. Helicobacter rodentii, Citrobacter rodenti and Corynebacterium bovis were all detected positive in different grades of animals. The positive rate of Helicobacter and Pseudomonas aeruginosa showed a downward trend, while Citrobacter rodentii, Corynebacterium bovis, Klebsiella pneumoniae The positive rate is on the rise.

  Conclusion: This study comprehensively and systematically analyzed the bacterial carrying status of experimental rats and mice in Shanghai, which played a role in improving the quality of experimental animals and the level of feeding and management, and provided a basis and reference for the revision and improvement of national standards for experimental animals in my country.
