Objective: To detect the expression of lipoic acid synthase (LIAS) in liver and kidney of leptin receptor-deficient Leprdb/db mice.
METHODS: Eight 10-week-old Leprdb/+ and Leprdb/db male mice were selected respectively. After fasting for 8 hours, the body weight and fasting blood glucose (FPG) of the two groups of mice were measured; the animals were anesthetized with ether, and blood was collected from the abdominal aorta. The animals were sacrificed, and the liver and kidney were collected and weighed. The right lobe and left kidney of the liver were fixed with 4% paraformaldehyde, and the histopathological examination of the liver and kidney was performed. LDL content. Mitochondria from liver and kidney tissue were isolated by mitochondrial isolation kit, total protein was extracted, and the expression of LIAS protein was detected by western blot method.
Results: Histopathological observation showed that the liver and kidney structure of Leprdb/ + mice were intact, while the liver cells of Leprdb/db mice showed steatosis, glomerular hypertrophy, thickening of the basement membrane, and widening of the mesangial area; Compared with mice, the body weight, GLU, CHO, TG, LDL and AST of Leprdb/db mice were significantly increased, and the difference was significant (P<0.05). The protein expression levels were all increased, and the difference was significant (P<0.05).
Conclusion: Leprdb/db mice with leptin receptor deficiency have glucose and lipid metabolism disorders, liver and kidney cell damage, and increased mitochondrial LIAS protein expression in liver and kidney tissues.
