OBJECTIVE: To screen polymorphic microsatellite markers in guinea pigs and provide basic information for identifying the genetic structure and gene location of guinea pigs.
Methods: The guinea pig genomic DNA data were screened from foreign websites, and the microsatellite sequences with AC and GT as the core were selected, and 400 pairs of primers were designed by software according to their flanking sequences. After two rounds of PCR and electrophoresis experiments, the electrophoresis bands were used as genetic markers, and the polymorphic primers were screened out by evaluating the structure of guinea pig microsatellites.
Result: In the first step, 10 DNA samples from 5 strains of guinea pigs were used for PCR, and about 110 pairs of guinea pig primers with polymorphism were screened from 400 pairs of primers, and the number of electrophoretic bands of the products was 1 to 3. In the second step, 60 DNA samples of 5 strains of guinea pigs were used for PCR, and 51 pairs of polymorphic primers with high electrophoresis band resolution were screened from the above 110 pairs of primers. Among them, more than 10 pairs of primers showed consistent markers or Mostly consistent differences.
Conclusion: The 51 pairs of primers screened are polymorphic among individuals and strains, and no other reports have been reported. They can be used for routine detection and research, and some of them may be used for guinea pig trait gene mapping.
