OBJECTIVE: To establish an in vitro model of intestinal epithelial cells—small intestinal organoid culture system, and to explore its related pathological detection techniques, so as to provide a convenient platform for the in vitro research of intestinal-related diseases.
Methods: Mouse intestinal epithelial crypts were isolated and cultured into small intestinal organoids to simulate the growth and development process of intestinal epithelium in vitro. By making paraffin sections, we explored the application of immunohistochemical techniques and three-dimensional immunofluorescence techniques based on organoids in Matrigel to detect the corresponding proliferation and differentiation signals.
RESULTS: The in vitro culture system of small intestinal organoids was explored and established. The application of paraffin section immunohistochemical technology and three-dimensional horizontal immunofluorescence technology can accurately detect the growth and development status of small intestinal epithelial structures.
Conclusion: The establishment of the in vitro culture system of small intestinal organoids and the application of immune detection technology will gradually make it the most favorable technical means for people to study intestinal related diseases.
