Objective: To test the tumorigenicity of High Five insect cell line to BALB/c nude mice, so as to ensure its safety as a vaccine production cell line.
Methods: Nude mice were randomly divided into 5 groups, namely High Five basic cell bank cell test group, High Five highest restricted passage cell test group, HEp-2 cell positive control group, CEF cell negative control group and no treatment. In the blank control group, nude mice were subcutaneously inoculated with the cell suspension on the back. After 3 weeks and 12 weeks, the nude mice were dissected and examined histopathologically for tumor formation.
Results: Nude mice in the positive control HEp-2 cell group formed nodules at the injection site 3 weeks after inoculation, and histopathological examination was squamous cell carcinoma; no nodules were formed in the negative control CEF cell group and High Five cell group, 12 days after inoculation. No tumor formation was found in Zhou's autopsy and histopathological examination.
CONCLUSION: Both the basic passage and the highest restriction passage High Five cells are not tumorigenic and can be safely used in vaccine production.
