OBJECTIVE: To analyze the expression levels of Rxra gene in 6 tissues of diabetic gerbils, in order to explore the molecular mechanism of diabetes in gerbils.
Methods: 6 diabetic gerbils and 6 normal gerbils were selected, and the skeletal muscle, liver, fat, kidney, heart and brain tissues of the animals were collected respectively. Real-time PCR and Western blotting were used to detect the mRNA of Rxra gene in each tissue. and protein expression levels.
RESULTS: The results of Real-time PCR showed that the mRNA level of Rxra gene was significantly decreased in the skeletal muscle and fat of the diabetic animals, and had no difference in the liver, while there was a trend of high expression in the kidney, heart and brain tissues. Western blotting results showed that the protein level of RXRA in the diabetic group was significantly decreased in skeletal muscle tissue, almost undetectable in adipose tissue, and the expression in other tissues was different, but there was no statistical difference.
Conclusion: The expression of Rxra is low in the skeletal muscle and adipose tissue of diabetic gerbils, and there is no difference in the expression levels in the other four tissues, indicating that the effect of Rxra on diabetes in gerbils mainly occurs in skeletal muscle and adipose tissue.
