Objective: To study the regulatory effect of mouse spleen mesenchymal stem cells (Sp-MSCs) on the proliferation and activation of T lymphocytes.
METHODS: Sp-MSCs were isolated and cultured from mouse spleen by tissue block method, and multi-directional differentiation experiments were performed. Mouse T lymphocytes were isolated, and lymphocyte transformation assay and mixed lymphocyte reaction were carried out to detect the effects of Sp-MSCs on T lymphocytes activated by non-specific antigens and alloantigens, respectively. The effect of Sp-MSCs on the proliferation of activated T lymphocytes was detected by CFSE flow cytometry. Quantitative PCR was used to analyze the effect of Sp-MSCs on inflammatory cytokines expressed by T lymphocytes.
RESULTS: The results of flow cytometry showed that Sp-MSsC could inhibit the proliferation of T lymphocytes induced by non-specific antigens and alloantigens. Sp-MSCs can effectively inhibit cell activation induced by lymphocyte transformation assay and mixed lymphocyte reaction. Further analysis found that Sp-MSCs inhibited T lymphocyte expression of interferon gamma, tumor necrosis factor alpha and interleukin 17A.
Conclusion: Sp-MSCs can inhibit the proliferation and activation of T lymphocytes, and inhibit the expression of various inflammatory cytokines in T lymphocytes.
