【Animal modeling】-In vitro differentiation of mouse umbilical cord mesenchymal stem cells

  Objective: To isolate and expand mouse umbilical cord mesenchymal stem cells to investigate whether they can be induced into cartilage, adipose and osteoblasts.

  Methods: The mUCMSCs were isolated, expanded and purified in vitro by adherent culture method, the morphological characteristics of the cells were observed under an inverted microscope, and the expression of antigen markers of the cells was detected and analyzed by flow cytometry. The isolated mUCMSCs were directionally induced and cultured into cartilage, adipose and osteoblast cells using the induction medium.

  RESULTS: Adherent fibroblast-like cells were isolated from fresh umbilical cord by tissue patch culture. These cells highly expressed CD29, CD90 and CD105, and low expressed CD34. After chondrogenesis induction, axin blue staining showed blue; after adipogenic induction, oil red O staining showed red lipid droplets; alizarin staining of osteogenic mUCMSCs showed red nodules.

  Conclusion: The mUCMSCs isolated and cultured by the adherent culture method can be induced to differentiate into cartilage, adipose and osteoblasts in vitro.
