【Animal Modeling】-Necrotic cells reverse the imbalance of Treg/Th17 ratio in septic mice

  Objective: To study the effect of pre-infusion of necrotic cells on the balance of Treg/Th17 cells in a mouse model of sepsis.

  METHODS: Thirty-four C57BL/6 mice were randomly divided into 4 groups: normal control group (PBS group, n=5), sham-operated control group (Sham group, n=5), and sepsis group (CLP group, n=5). 12) and necrotic cell treatment group (Nec group, n=12). Normal mice in the PBS group were injected intraperitoneally with 200 μl PBS once. The mice in the Sham group were operated to separate the end of the cecum without ligation and perforation; the mice in the CLP group were treated with cecal ligation and perforation (CLP) to establish a sepsis model; The mice were pre-injected intraperitoneally with 2×107 necrotic cells, and the cecum was ligated and perforated on the 5th day. All mice were sacrificed 2 weeks after the preparation of the CLP model, and the proportions of CD4+Foxp3+ Treg cells and CD4+IL17A+ Th17 cells in peripheral blood, spleen and thymus were detected by flow cytometry.

  Results: Th17 ratio and Treg/Th17 ratio in spleen of CLP group were significantly higher than those of PBS group and sham group (P<0.01); Injection of necrotic cells could reverse the imbalance of Th17 ratio, Treg ratio and Treg/Th17 ratio in mice after CLP surgery (P<0.05).

  Conclusion: Pre-infusion of necrotic cells can reverse the imbalance of Treg/Th17 cell subsets in septic mice.
