【Animal Modeling】-Interventional effect of flavonoids in Sandianjin on ulcerative colitis mice and their effects on intestinal flora

  Objective To investigate the intervention effect of the national medicine Sandianjin total flavonoids and its monomer compounds vervasoflavone and formononetin on dextran sodium sulfate (DSS)-induced ulcerative colitis (UC) mice. Influence of gut microbiota.

  Methods The UC mouse model was established with 3% DSS, and 30 C57BL/6 mice were divided into normal group, model group, total flavonoids group (500 mg/kg), mullein group (100 mg/kg) and Formononetin group (100 mg/kg). The weight changes of mice were observed, disease activity index (DAI) and colon histopathology were scored, and the levels of IL-1β and IL-6 in the serum of mice were detected, and the changes in the intestinal flora of mice were analyzed by high-throughput sequencing.

  Results The total flavonoids, isoflavones and formononetin of Sanjinjin can effectively alleviate the symptoms of colitis in mice and reduce the expression of inflammatory factors. The diversity of tract flora increased significantly, and at the family level, the proportion of pathogenic bacteria Helicobacter decreased, and the proportion of Muribaculaceae and Lachnospira increased. The reduction of Bacteroidetes; the reduction of opportunistic pathogens Bacteroides and Escherichia coli at the species level.

  Conclusion Three-point gold total flavonoids, isoflavones and formononetin can improve the signs and inflammatory factors of UC mice to a certain extent, and have a good recovery effect on intestinal flora.
