Objective To explore the effect of Bushen Yiqi Huayu Granule on angiogenesis in osteoporosis rats based on TLRs/NF-κB signaling pathway.
Methods Sixty SPF female SD rats were randomly divided into a sham operation group, a model group, a granule group and a positive control group, 15 rats in each group. Except for the sham operation group, the other three groups underwent bilateral ovariectomy for bone preparation. The sham operation group only underwent surgery without removing the ovaries as a control. Rats in the granule group were given 5 g/kg Bushen Yiqi Huayu granules by gavage, while the rats in the positive control group were given 0.5 mg/kg sodium alendronate by gavage. The rats in the model group and the sham-operated group were given An equal volume of normal saline was given by gavage. The rats were sacrificed after continuous administration for 12 weeks, and the femur tissue of the rats was subjected to bone mineral density detection, HE staining observation, Micro-CT three-dimensional reconstruction analysis, CD31 fluorescence immunoassay and blood perfusion imaging analysis; ELISA was used to detect VEGF and HIF-1α in serum. concentration; Western blot was used to detect the expression of p50/p-p50 and p65/p-p65 proteins.
Results Compared with the sham-operated group, the model group had lower bone mineral density (P<0.05), sparse bone trabecular growth, slower blood vessel formation, and fewer blood vessels, and the serum VEGF and HIF-1α concentrations were significantly decreased (P<0.05). 0.05), p50/p-p50 and p65/p-p65 protein expressions in femoral tissue were significantly increased; compared with the model group, the bone trabeculae of the granule group and the positive control group grew well, and the number of blood vessels formed was more. The bone mineral density was significantly increased, the serum VEGF and HIF-1α concentrations were significantly increased (P<0.05), and the p50/p-p50 and p65/p-p65 protein expressions were significantly decreased (P<0.05).
Conclusion Bushen Yiqi Huayu Granule may promote the formation of blood vessels in osteoporotic rats by regulating TLRs/NF-κB signaling pathway, and play an important role in relieving osteoporosis.
