Objective To explore the effect and mechanism of Yanlishuangkouhan dropping pills on chronic pharyngitis rabbit model.
Methods Forty-two healthy New Zealand white rabbits were randomly divided into 7 groups: blank group, model group, Manyanshuningqinghouliyan granules group (9 g/kg), Yanlishuangkouhan dropping pills low (0.025 g/kg), medium ( 0.05 g/kg), high (0.1 g/kg) dose group and nebulized group (0.025 g/kg). Except for the blank group, the other rabbits were used to establish the animal model of chronic pharyngitis by chemical stimulation. From the 2nd day after the successful modeling, the 5 treatment groups were treated with the corresponding doses of Manyan Shuning Qinghou Liyan Granules and Yanli Shuangkou Han Dropping Pills, respectively, while the blank group and model group were treated with sucrose (0.025 g/kg) It was placed in the oral cavity of rabbits and administered continuously for 7 days. From the 3rd day of modeling, the local condition of the throat wall of the rabbits was observed every day, blood was collected after the administration, and the rabbits were sacrificed for sampling for examination. Histopathological changes of the throat wall mucosa were observed by HE staining; CD3+, CD4+, CD8+ were detected by flow cytometry, and CD4+/CD8+ levels were calculated; serum interleukin-6 (IL-6), interleukin-1 (IL-1) were detected by ELISA -1), tumor necrosis factor-α (TNF-α), anti-C
Responsive protein (CRP) and total antioxidant capacity (T-AOC) levels; Western blotting was used to detect IL-6, IL-1, TNF-α, nuclear transcription factor κB (NF-κB p65), nuclear transcription Factor-κB inhibitory factor (IκB-α) protein expression; real-time fluorescence quantitative PCR detection of NF-κB p65 and IκB-α mRNA expression in the mucous membrane tissue of the throat wall.
Results Compared with the blank group, the laryngopharyngeal wall of rabbits with chronic pharyngitis
Mucosal pathological tissue damage; the levels of CD3+, CD4+, CD4+/CD8+ and T-AOC were significantly decreased, and the levels of CD8+, IL-6, IL-1, TNF-α, CRP, NF-κB p65 and IκB-α were significantly increased (P<0.01). Compared with the model group, different treatment groups can reduce the pathological damage of the throat wall mucosa to different degrees; the high-dose Yanlishuangkou Handropping group and the atomization group significantly increased the levels of CD3+, CD4+, CD4+/CD8+ and T-AOC, and CD8+ , IL-6, IL-1, TNF-α, CRP, NF-κB p65 and IκB-α levels were decreased (P < 0.05), and the effect was better than the low and medium doses and the slow Yan Shuning Qinghou Liyan granules groups.
Conclusion Yanli Shuangkou Han-Dropping Pills can significantly improve the inflammatory indexes of the chronic pharyngitis rabbit model, reduce the histopathological damage, and regulate the expression of NF-κB signaling pathway-related factors, and the effect of atomization is more obvious.
