【Animal modeling】-Effect of Naoshuantong capsule on the expression of inflammatory factors in aorta of ApoE-/- mice

Objective To observe the effect of Naoshuantong capsule on the expression of inflammatory related factors interleukin-1β, interleukin-6, tumor necrosis factor, intercellular adhesion molecule-1 and metalloproteinase-9 in ApoE-/- atherosclerosis mouse model. The mechanism of its treatment of atherosclerosis was preliminarily discussed.

Methods 8-week-old male ApoE-/- mice were given a high-fat diet to establish an atherosclerosis model. After successful modeling, they were randomly divided into a model control group, a low-dose Naoshuantong capsule group, and a high-dose Naoshuantong capsule group. . The low-dose group was given Naoshuantong capsule 0.5 g/kg body weight, the high-dose group was given Naoshuantong capsule 1.0 g/kg body weight, and the model control group was given the same amount of distilled water. After continuous administration for 12 weeks, the whole aortic intimal plaque was detected by red oil O staining, and the inflammatory related factors IL-1β, IL-6, TNF-α, ICAM-1 and MMP-9 were observed and compared in each group of mice. Express.

Results After drug intervention, the aortic plaque area in the high-dose Naoshuantong capsule group was significantly lower than that in the model control group (P<0.01). Compared with the model control group, the low-dose Naoshuantong Capsule group could inhibit the protein expression levels of IL-1β and IL-6 related to inflammation respectively (P<0.05), and the IL-1β in the Naoshuantong high-dose group , IL-6, TNF-α and ICAM-1 expressions were decreased (P<0.05, P<0.mmp-9="" p="">0.05).

Conclusion Naoshuantong Capsules can regulate the expression levels of inflammatory-related factors, and its mechanism of action in the treatment of atherosclerosis is likely to be related to this pathway.
