【Animal Modeling】-Influence of the placement time of venous blood cell suspension on the determination of blood routine in mice

  Objective To observe the effect of placing time of venous blood cell suspension on blood routine determination in mice.

  Methods 20 adult ICR mice and 20 C57BL/6J mice were randomly selected. Quantitative venous blood was collected and added to the diluent for quantitative blood cell analysis. , 4h, .6h, 8h in MEK-6410C automatic blood cell analyzer using micro-blood assay to determine blood routine.

  Results The changes of blood routine data of ICR mice and C57BL/6J mice were basically the same. Compared with 0min, RBC was significantly decreased at 2h, 4h, 6h and 8h (P<0.01), PLT was significantly increased at 4h, 6h and 8h (P<0.01), and PCT was significantly increased at 2h, 4h, 6h and 8h. Significantly increased (P<0.01), MPV was significantly increased at 2h, 4h, 6h and 8h (P<0.01), MCV was significantly decreased at 1h, 2h, 4h, 6h and 8h (P<0.01), MCHC Significantly increased at 30min, 1h, 2h, 4h, 6h and 8h (P<0.01), MCH increased significantly at 1h, 2h, 4h, 6h and 8h (P<0.01), HCT at 1h, 2h , 4h, 6h and 8h were significantly decreased (P<0.01), while="" pdw="" and="" hgb="" had="" no="" significant="" change="" p="">0.05).

  Conclusion When the blood routine of mice is determined by microblood assay, it should be done as soon as possible after the venous blood cell suspension is prepared. The total number of RBCs should be measured within 1 hour, the determination of HCT and MCV indicators related to RBC should not exceed 30 minutes, the determination of PLT should be completed within 2 hours, and the determination of PCT and MPV related to PLT should be within 1 hour. completed within 8 hours.
