【Animal Modeling】-Study on the effect of vepromide combined with fluoxetine hydrochloride on depressive mice

  Objective To investigate the effect of Weipronamine capsules combined with fluoxetine hydrochloride capsules on depression in mice.

  Methods The mouse model of depression was established by the method of chronic stress and solitary rearing. Randomly divided into 5 groups: blank control group, model group, retinol group, fluoxetine hydrochloride group and revetamine combined with fluoxetine hydrochloride group (combination group), 10 animals in each group, and were given intragastric administration for 30 consecutive days. The changes in body weight, sugar water preference rate, forced swimming immobility time and tail suspension immobility time of mice in each group were observed for comparative analysis, and the effect of combined medication on depressed mice was comprehensively evaluated.

  Results When the modeling was completed (the 0th day of administration), compared with the blank control group, the total consumption of sugar water, the preference rate of sugar water and body weight were significantly decreased in each depression model group (P<0.01). After intragastric administration, the body weight of animals in each experimental group increased. On the 30th day, compared with the model group, there was a significant difference in the weight increase of the combined drug group (P<0.05). On the 39th day, the vipronamine group There was a significant difference in weight gain (P<0.05). On the 46th day, there was a significant difference in weight gain between the combined administration group and the fluoxetine hydrochloride group (P<0.05). On the 50th day, the total consumption of sugar water and the preference rate of sugar water in each experimental group increased to varying degrees. Compared with the model group, there were significant differences in the total consumption of sugar water and the preference rate of sugar water in the Vipro group (P<0.05). , compared with the model group, there was a significant difference in the total consumption of sugar and water in the combination group (P<0.05), and there was a very significant difference in the preference rate of sugar and water in the combination group (P<0.01). The immobility time of forced swimming and the immobility time of tail suspension in the retinamine group and the combined drug group were significantly reduced. Compared with the model group, there were significant differences in the immobility time of swimming and tail suspension in the retinamine group (P<0.05). There was a very significant difference in the immobility time of tail suspension (P<0.01). Compared with the fluoxetine hydrochloride group, there was a significant difference in the immobility time of tail suspension in the combination group (P<0.05).

  Conclusion The experiments suggest that viprotamine has antidepressant effect, and the combined effect of fluoxetine hydrochloride is better. This study will provide experimental basis and reference for clinical medication of clinical depression.
