Objective To investigate the effect of miR-124-3p on the proliferation and invasion of placental trophoblast cells in preeclamptic rats by targeting MAPK 14.
Methods The placental trophoblast tissue and primary cells of normal rats and model rats were extracted and transfected in groups. MTT, flow cytometry and Transwell were used to detect cell viability, apoptosis, cycle, migration and invasion ability, and qRT-PCR and Western blot to detect the expression levels of factors. The targeting relationship between miR-124-3p and MAPK 14 was verified.
Results After overexpression of miR-124-3p, the cell proliferation and invasion ability decreased, the cells were arrested in the G1/G0 phase, and the apoptosis increased; after inhibiting the expression of miR-124-3p, the results were opposite. The dual-luciferase reporter assay showed that miR-124-3p had a targeting relationship with MAPK 14. After inhibiting the expression of MAPK 14, the results were consistent with the results of over-expression of miR-124-3p, the cell viability and invasion ability were decreased, and the apoptosis was increased; the experimental group of over-expression of MAPK 14 was consistent with the results of inhibiting the expression of miR-124-3p.
Conclusion miR-124-3p can target and negatively regulate MAPK 14, and have an effect on the proliferation and invasion of placental trophoblast cells in preeclamptic rats.
