【Animal modeling】-Effect of Fujiutie ointment on the expression of NGF/TrKA and related inflammatory factors in asthmatic mice

  Objective: To observe the effect of the preparation "Fujiu Tie Gao" of the Affiliated Hospital of Liaoning University of Traditional Chinese Medicine on serum interleukin-4, interferon γ, related immunoglobulin E cytokines, nerve growth factor and tyrosine kinase A protein in the lung tissue of asthmatic mice. To explore the effect of its expression on the treatment of asthma.

  Methods: Fifty adult BALB/c mice were randomly divided into 5 groups: A normal control group, B asthma model group, C dexamethasone group, D Fujiutie high-dose group, and E Fujiutie low-dose group. Ovalbumin (ovalbumin, OVA) was sensitized by intraperitoneal injection and challenged by ultrasonic aerosol inhalation to establish an asthma model. The expressions of IL-4, total IGE, OVA-specific IGE and IFN-γ cytokines in serum were detected by ELISA, HE staining and Masson staining The pathological changes were observed, and the expression of NGF and TrKA protein in lung tissue was observed by immunohistochemistry.

  Results: Compared with the normal group, the expression of NGF, TrkA and IL-4, total IGE and OVA-specific IGE in the model group were significantly increased (P<0.05), and the expression of IFN-γ was decreased (P<0.05); Compared with the dexamethasone group and the Fujiutie high-dose group, the expressions of NGF, TrkA, IL-4, total IGE, and OVA-specific IGE were decreased (P<0.05), and the expression of IFN-γ was increased (P<0.05). volt="" p="">0.05).

  Conclusion: Fujiu plaster can inhibit the secretion of IL-4 and IGE by down-regulating the expression of NGF and TrKA proteins, and at the same time promote the secretion of IFN-γ, so that TH1 and TH2 tend to balance, thereby relieving asthma attacks.
