Objective To observe the anesthesia effect and internal environment stability of mice receiving midazolam combined with butorphanol before isoflurane anesthesia, and evaluate the safety.
Methods A total of 75 mice were selected and randomly divided into 3 groups: control group, mice were only given isoflurane anesthesia; observation group, mice were given midazolam combined with butorphanol before isoflurane anesthesia; In the blank group, the mice were not given any anesthesia treatment. 25 per group. Heart rate and blood oxygen saturation (SpO2) were measured immediately after induction and every 5 min within 1 h after induction. The serum blood glucose (Glu) and cortisol (Cor) levels of mice in each group were compared before and during anesthesia. The differences in the induction and recovery time, analgesic and sedative effects of the mice in each group were compared. The differences in the incidence of adverse reactions in each group during the induction process were compared.
Results Compared with the control group, the induction time, sedation onset time and pain disappearance time in the observation group were significantly lower (P<0.05), but="" there="" was="" no="" difference="" in="" the="" recovery="" time="" p="">0.05). The heart rate of the blank group was higher than that of the control group and the observation group (P<0.05); 5-35 min after induction, the heart rate of the control group was higher than that of the observation group (P<0.05); 35-60 min after the induction, the heart rate of the control group was lower than that of the control group (P<0.05). Observation group (P<0.05). 1h after induction, the SpO2 of the control group was lower than that of the observation group and the blank group (P<0.spo2="" p="">0.05). The changing trend of heart rate among the three groups was different (F = 15.215, P = 0.000). There was no difference in the changing trend of SpO2 among the three groups (F= 1.015, P= 0.180). Before anesthesia, there was no significant difference in the levels of Cor and Glu among the three groups (P>0.05); during anesthesia, the levels of Cor and Glu in the control group were higher than those in the blank group and the observation group (P<0.05), while="" the="" blank="" group="" and="" observation="" glu="" p="">0.05). The intraoperative Cor and Glu of the control group were higher than those before operation (P<0.05), while="" the="" blank="" group="" and="" observation="" were="" glu="" p="">0.05). There was no significant difference in the total incidence of adverse reactions between the control group and the observation group (P=0.626).
Conclusions The anesthesia effect of midazolam combined with butorphanol before isoflurane anesthesia is reliable, the induction of anesthesia is stable, the respiratory depression is relieved, and the safety is high.
