Objective The purpose of this experiment was to study the anti-tumor effect of Xiuzhen Mushroom and its anti-tumor mechanism.
Methods 60 ICR male mice were selected, 10 mice were selected as blank group, and the remaining 50 mice were inoculated with H22 mouse ascites subcutaneously in the armpit to establish a tumor-bearing mouse model. After modeling, they were randomly divided into model group, positive drug (CTX ) group and Xiuzhen mushroom low, medium and high dose groups, 10 in each group. The mice in the model group were given normal saline, the mice in the positive drug group were injected with cyclophosphamide (CTX) normal saline solution by intraperitoneal injection at 20 mg/kg body weight every other day, and the mice in the low, medium and high dose groups of Xiuzhen mushroom were given 750, 1500, 3000 mg/kg body weight daily doses of Xiuzhen Mushroom physiological saline suspension. After 10 days of administration, the average tumor weight and tumor inhibition rate of the mice in each group were determined; the index of immune organs, serum immunoglobulin and cytokine contents were determined; the antioxidant indexes of liver and kidney were determined; the HE staining of tumor and spleen tissue was observed. pathological section.
Results (1) The average tumor weight of mice in the low, medium and high dose groups of CTX and Xiuzhen mushroom was significantly lower than that in the model group (P<0.01). The tumor inhibition rates of the four groups were 55.18%, 29. 06%, 47. 47% and 48. 80%.
(2) Compared with the blank group, the spleen index, serum IgA and TNF-α contents and liver MDA contents of mice in the model group were significantly increased (P<0.05, P<0.01), and the serum IL-6 content increased significantly (P<0.05, P<0.01). trend="" p="">0. 05), while serum IL-2 content, liver CAT and GSH-Px activities and renal SOD, GSH-Px and CAT activities were significantly decreased (P < 0. 05, P < 0. 01); , the thymus index of mice in CTX group was significantly lower than that in blank group (P<0.05).
(3) Xiuzhen Mushroom has a reversal effect on the abnormal changes of immune and antioxidant functions in tumor-bearing mice: compared with the model group, the levels of serum IgA and TNF-α in mice in each dose group of Xiuzhen mushroom and the level of IL-6 in mice in high dose group significantly decreased (P < 0. 05, P < 0. 01), and the serum IL-2 level of the mice in the low-dose group was significantly increased (P < 0. 01); The activity of CAT in the liver and the activities of SOD and CAT in the kidney (P < 0. 05, P < 0. 01) significantly increased the activity of liver GSH-Px in the middle and high dose groups and the GSH-Px activity in the kidney in the low and middle dose groups (P < 0. 01). 0. 05, P < 0. 01), and significantly decreased the hepatic MDA content in the middle-dose group (P < 0. 05).
(4) Compared with the CTX group, the spleen index of the mice in the Xiuzhen mushroom high-dose group was significantly increased (P < 0. 05), while the serum IL-6 level was significantly decreased (P < 0. 05).
(5) The pathological sections of tumor tissue showed that the tumor necrosis area increased significantly in the CTX group and each dose group of Xiuzhen mushroom.
Conclusion Xiuzhen Mushroom can inhibit the growth of H22 solid xenograft tumor, and the mechanism is related to its strong immune regulation and antioxidant effects.
