Objective: In order to explore the stable, reliable and reasonable fasting time of OGTT in Kunming mice, which are used the most in China in a specific environment.
Methods: Oral glucose tolerance test (OGTT) in mice is a method to study the function of islet β cells and the ability of blood sugar regulation. Fasting before the experiment is a necessary step, but under different fasting times, the mice's There are certain differences in fasting and glucose-stimulated blood glucose and insulin concentrations. In this experiment, clean-grade Kunming male mice aged 6 to 7 weeks were selected and randomly divided into four groups. OGTT was performed after fasting for 4, 8, 10, and 12 hours respectively. , detect the blood glucose concentration and insulin concentration of mice in each group, and analyze the changing trend of blood glucose concentration and insulin concentration.
Results: (1) The blood glucose concentration and insulin concentration of all mice increased from the fasting reference value to the critical value and then dropped to the reference value. Compared with the fasting blood glucose concentration of the 4-h fasted mice, the fasting blood glucose concentration of the other three groups of mice The average blood glucose concentration was significantly lower than that of the fasting 4h group (P<0.01). (2) From the point of view of blood sugar, the fasting 10h group had the largest increase in the blood glucose concentration from the fasting critical value. Compared with the fasting group for 4 hours, there was a significant difference (P<0.05). (3) From the perspective of insulin secretion, the fasting insulin concentration of fasting for 4 hours was 4% higher than that of fasting for 8 hours, which was about twice that of fasting insulin for 12 hours. The area under the insulin curve was the smallest at 8 h after eating.
Conclusion: Kunming mice fasted for 8-10 hours in the OGTT, the blood glucose concentration and insulin concentration have large differences, high resolution and good change trend; the arrangement of fasting time is more reasonable, and the experimental plan is reasonable and efficient. When establishing a diabetic model in mice, the fasting time of the glucose tolerance test can be used as a reference.
