【Animal modeling】-Efficacy observation and action pathway of Eucommia ulmoides extract on lipopolysaccharide-induced mastitis mice

  Objective: To study the therapeutic effect and mechanism of Eucommia ulmoides extract on mastitis in mice.

  Methods: A mouse model of mastitis was established by perfusing LPS into the milk duct. After 12 hours of LPS injection, different concentration gradients of Eucommia were administered through the abdominal cavity, and the mice were divided into 6 groups. The pathological changes of each group were observed by staining, and the changes of TNF-2α, IL-6 and IL-1β cytokines in breast tissue were detected by ELISA.

  Results: 1) LPS at a concentration of 50 μL of 0.2 mg/mL could successfully build a mouse mastitis model; (2) HE staining results showed that Eucommia ulmoides extract could reduce the degree of injury of LPS-induced mouse mastitis. With the increase of the dose of eucommia ulmoides, the damage degree of breast tissue was alleviated, and the injection dose was 20 mg/kg and 27 mg/kg. The content of IL-6 and TNF-α, and the injection dose of 27 mg/kg had the best effect (P<0.01).

  Conclusion: Eucommia ulmoides extract can significantly reduce the pathological damage of mouse mammary tissue, reduce the release of inflammatory cytokines IL-1β, IL-6 and TNF-α in the mammary tissue, and block the neutrophils in the mammary tissue and mammary gland. The recruitment and migration process of alveolar cavity can reduce the damage of breast tissue, and the anti-inflammatory effect of Eucommia ulmoides extract at a concentration of 27 mg/kg is the best.
