Purpose requirements
1. Understanding mouse peritoneal macrophages
2. The principle of phagocytosis. Familiar with the basic process of phagocytosis
Fundamental
Cell phagocytosis is a method of obtaining nutrients from unicellular animals. Macrophages, monocytes and neutrophils in higher animals have phagocytosis. It is an important part of the human body's non-specific immune function. Macrophages are produced by differentiation of bone marrow stem cells. When pathogenic microorganisms and other foreign bodies enter the human body, macrophages accumulate in the foreign body due to their chemotaxis. Macrophages bring them into the cytoplasm and form phagocyte vesicles. Then, it fuses with lysosomes inside the cell to digest and break down foreign bodies. The formation of phagocyte vesicles requires the help of microfilaments and their binding proteins. If cells are treated with cytochalasin B, a drug that degrades microfilaments, it prevents the formation of phagocytic vesicles, and the immunosuppressant cyclophosphamide (CYP) may affect macrophages. The phagocytosis is also affected.
laboratory supplies
1. Equipment: microscope, dissection board, scissors, tweezers, syringe, glass slide, cover glass, pipette, etc.
2. Reagent: 0.85% saline; Alsever blood preservation solution; 6.0 g of water-soluble starch and 100 ml of water are boiled for use.
3. Material: mouse, 1% chicken red blood cell suspension
Methods and steps
Collect venous blood, add saline and centrifuge twice, depending on the number of red blood cells obtained. Adjust the red blood cell concentration of chickens by adding a certain amount of saline to 1%
