Objective: To investigate the inhibition of proliferation and induction of apoptosis of human colon cancer HT29 cells by deoxyvioletine and its possible mechanism.
Methods: HT29 cells were treated with different concentrations of deoxyvioletine, and the tetramethylazolium blue (MTT) colorimetric method was used to detect the inhibition rate of deoxyvioletin on HT29 cells; flow cytometry was used to detect cell cycle and apoptosis. apoptosis; Western Blot method was used to detect the changes of apoptosis-related proteins Bax, Bcl-2 and Caspase9 expression.
Results: Deoxyvioletine had obvious inhibitory effect on the proliferation of HT29 cells; it could increase the proportion of cells in G0/G1 phase and decrease the proportion of cells in S phase; increased apoptosis rate, increased protein expression of Bax and Caspase9, and increased protein Bcl-2 expression decreased.
Conclusion: Deoxyvioletin inhibits the proliferation and induces apoptosis of colon cancer HT29 cells, and its apoptosis mechanism may act on the mitochondria-mediated endogenous pathway.
