【Animal modeling】-Effect of low selenium diet on the expressions of CD163, CD206 and CCR7 in the liver and kidney of rats

  Objective: To investigate the effect of low selenium diet on liver and kidney in rats, and the role of macrophage polarization to M1 and M2 in liver and kidney fibrosis.

  METHODS: Rats were fed with low-selenium and normal diets to establish a low-selenium animal model. Twelve female and 12 male SD rats were divided into 2 groups according to the random number table method: control group (group Z, feed selenium 0.18 mg/kg), low selenium group (group S, feed selenium 0.02 mg/kg), feeding After 109 days, the rats were sacrificed, and the pathological changes of the liver and kidney were observed by HE staining, and the number of CCR7, CD206, and CD163 positive cells in the liver and kidney tissue was detected by immunohistochemistry using SP staining.

  Results: (1) The liver and kidney fibrosis of the rats in the low-selenium group was more serious than that in the control group; (2) The staining intensity of CCR7 and CD206 in the low-selenium liver group of male and female rats was significantly higher than that of the control group (female, male, female, male, P>0.05. ) were not statistically different. The staining intensity of CD163 was lower than that of the control group (P<0.05 for both females and males), which was statistically significant; (3) The staining intensity of ccr7 in the proximal tubules of the low-selenium group in the kidneys of male and female rats was slightly higher than that of the control group (P<0.05 for="" both="" females="" and="" .="">0.05), slightly lower in the distal tubule than the control group (female P>0.05, male P<0.05), but the results were not statistically different; CD163 staining intensity in the distal tubule was lower than that in the control group (female, male P<0.05) P<0.05), the staining intensity of CD206 in the proximal tubule was higher than that in the control group (P<0.05 for="" females="" and="" which="" was="" statistically="" the="" staining="" intensity="" of="" cd163="" in="" proximal="" tubule="" lower="" than="" that="" control="" group="" p="">0.05 for females , male P<0.05), females had no statistical significance, males had statistical significance, CD206 staining intensity in distal tubules was higher than that of the control group (females P<0.05, males="" p="">0.05), females had statistical significance, males had no Statistical significance.

  Conclusion: The liver and kidney of rats in low selenium group had obvious fibrosis, and low selenium might inhibit the polarization of macrophages to M2 type.
