Objective: To study the toxic effects of doxorubicin liposome injection and doxorubicin injection by continuous intravenous injection on rats and compare the similarities and differences of toxicity.
Methods: Ninety SD rats were randomly divided into solvent control group, doxorubicin liposome injection (1 mg/kg) and doxorubicin injection (1 mg/kg) groups. Rats in each group were intravenously injected once every 3 days for 13 consecutive times, and their body weight, blood biochemistry, hematology, organ coefficient and other indicators were detected, and histopathological examinations were performed.
RESULTS: Doxorubicin liposome injection 1 mg/kg could induce hair loss and ulcers in SD rats, significantly delayed body weight gain, increased PLT, ALT, BUN, decreased ALB, increased cardiac and renal coefficients, increased thymus and testis The coefficient decreased, and the results of histopathological examination showed that the seminiferous tubules of the testis were sloughed off, the spermatogenic cells at all levels disappeared, and some interstitial edema; cardiac myocardial fibers were ruptured, dissolved, and striated and some nuclei disappeared; renal tubules appeared in the kidneys. Histopathological lesions such as hyaline casts. Compared with doxorubicin liposome injection, the same dose of doxorubicin injection can also cause the above lesions, but the rate of hair loss and skin ulcer in rats decreased, and the renal toxicity and cardiotoxicity were significantly increased.
Conclusion: Compared with ordinary injection, doxorubicin liposome injection has significantly reduced cardiac and renal toxicity, but significantly increased skin toxicity.
