【Animal Modeling】-Effects of hesperidin on antioxidant activity and gene expression of antioxidant enzymes in mice

  Objective: To investigate the effect of hesperidin (HDN) on the antioxidant effect of the body.

  Methods: Spectrophotometry, pyrogallol auto-oxidation method and Fe2+-phenanthroline method were used to detect HDN scavenging free radicals, inhibiting mitochondrial swelling and oxidative hemolysis of red blood cells in vitro; experimental mice were fed with different concentrations of HDN (0, 80, 160, 320 mg/kg) for 12 consecutive days, ELISA and spectrophotometry were used to detect the content of MDA, the activity of antioxidant enzymes (SOD, CAT, GSH-PX) in mouse tissues, and the expression level of antioxidant enzymes mRNA was analyzed by RT-PCR. .

  Results: Compared with the control group, the scavenging rate of free radicals (·OH, O2-·, DPPH·) in the HDN group was significantly increased, and the oxidative hemolysis and mitochondrial swelling of mouse erythrocytes in vitro were significantly reduced; The activities of oxidase SOD, CAT and GSH-PX were significantly higher than those of the control group, and the expression of antioxidant enzyme mRNA (SOD, CAT, GSH-PX) in tissues was up-regulated.

  Conclusion: HDN can scavenge free radicals, reduce the oxidative damage of cells caused by free radicals, inhibit the generation of peroxides, up-regulate the expression of antioxidant enzymes and improve the activity of enzymes, showing a good antioxidant effect.
