Objective: To study the effect of lentivirus-mediated survivin gene-specific short hairpin RNA (shRNA) on the growth of human endometrium in the peritoneal cavity of nude mice.
Methods: Forty-five nude mice were randomly divided into experimental group, negative control group and blank control group, 15 mice in each group. Virus, unloaded lentivirus and phosphate buffered saline were injected into the abdominal cavity of three groups of nude mice respectively. After 2 weeks, the success rate of endometrial implantation in each group was observed. The morphological characteristics of ectopic lesions were observed under light microscope, and immunohistochemical methods The expression levels of survivin in the lesions of each group were detected.
Results: The success rate of endometrial implantation in the experimental group (26.67%) was significantly lower than that in the negative control group (73.33%) and the blank control group (80%, P<0.01). The interstitium was accompanied by different degrees of necrosis; the expression of survivin protein in the lesions of this group was also significantly decreased, and the difference was significant compared with the two control groups (P<0.001).
Conclusion: Lentivirus-mediated survivin gene-specific shRNA can significantly inhibit the growth of human endometrium in the abdominal cavity of nude mice by targeting silencing of the survivin gene.
