【Animal modeling】-Arachidonic acid-induced establishment of diffuse cerebral thrombosis model in rats

  Objective To establish and optimize the rat model of diffuse cerebral thrombosis induced by arachidonic acid.

  Methods A rat model of diffuse cerebral thrombosis was established by injecting arachidonic acid through the internal carotid artery; meanwhile, a sham-operated group was set as a control. The modeling effect was evaluated by detecting the exudate content of Evans blue (EB) in brain tissue and detecting the degree of pathological damage in brain tissue by HE staining.

  Results Compared with the sham-operated group, diffuse EB exudation appeared in the injured side brain tissue of the model group (P<0.01), and the exudation was more uniform. The pathological manifestations of the injured side of the brain were disordered cell arrangement, shrinking nuclei, enlarged intercellular spaces, increased vacuolar cells, and individual microvessels with red strip-shaped ischemia pathological damage.

  Conclusion Arachidonic acid can successfully induce the diffuse cerebral thrombosis model, and the optimized method has better stability and lower mortality.
