Objective To study the modeling points of constipation animal models and provide reference for improving the success rate of modeling.
Methods Using "constipation" and "animal model" as the subject headings, all experimental journal literatures from January 2015 to July 2020 in CNKI, Wanfang and VIP databases were retrieved. The types of experimental animals, modeling methods, modeling methods, modeling time, and detection indicators were summarized and analyzed.
Results A total of 142 journal articles that met the criteria were included. The animal models of constipation were mainly replicated by male and female SD rats and KM mice. The most common modeling method was the compound diphenoxylate and loperamide gavage. The modeling time was mostly 7d and 14d, and the longest was 120d for slow transit constipation. The index detection focused on carbon ink propulsion rate, defecation function, colon tissue immunohistochemistry and serum biochemical indexes.
Conclusion The existing animal models are often modeled by drug-induced single factor, but lack of multi-factor modeling evaluation system, such as water-restricted diet method and low-fiber diet method are more consistent with the pathogenic factors of real constipation, but the current related research is not effective. At present, most studies on constipation models tend to evaluate the tested drugs, such as intestinal defecation function, colon histopathological structure and biochemical indicators in serum. With the deepening of constipation research, the interaction of brain-gut axis provides new ideas for constipation research, and related experimental research can be added in future constipation experiments.
