【Animal Modeling】-Application analysis of 274 cases of rheumatoid arthritis animal model based on data mining

  Objective To study the modeling elements of rheumatoid arthritis animal models, and to provide methodological reference for improving the success rate of modeling and evaluating the effectiveness of the tested drugs.

  Methods Using rheumatoid arthritis and animal models as the subject headings, the relevant journal literatures of China National Academic Journals Full-text Database (CNKI) and Wanfang Database (January 2017-June 2020) were searched, and the types of experimental animals, stimulation methods, and inflammatory conditions were collected. Methods, detection indicators, etc., establish a database, and use Excel 2013, SPSS Statistics 19.0 statistical software to perform association rule analysis and factor analysis on the imported detection indicators.

  Results 274 journal articles that met the criteria were included, among which the most used experimental animal species were male SD rats (69 times, 25.18%) and male Wistar rats (54 times, 19.71%); the most commonly used inflammatory methods were CIA method (172 times, 62.77%) and AA method (82 times, 29.93%), etc.; the most commonly used stimulation methods are subcutaneous injection at the base of the tail (71 times. 25.91%) and intradermal injection of the right hind foot plantar (52 The most frequently detected indicators were the degree of swelling of the soles of the feet (133 times, 17.48%), the arthritis score index (84 times, 11.04%), and the level of TNF-α in serum (74 times, 9.72%) and joint histopathology (66 times, 8.67%) and so on.

  Conclusion It is recommended to use male SD rats or male Wistar rats for the establishment of rheumatoid arthritis animal models. The inflammation is induced by subcutaneous injection at the base of the tail, and bovine type II collagen and Freund's incomplete or complete adjuvant 1∶1 are used. The mixed emulsion, 0.1 mL per point to induce inflammation, can improve the success rate of the model. After reviewing the literature for nearly three years, a total of 274 literatures that met the requirements were screened. This paper analyzes the existing animal models, and puts forward suggestions for the further improvement of rheumatoid arthritis animal models and the standardization of model evaluation, and to improve the consistency between animal models and clinical practice. Summarize the research progress of rheumatoid arthritis animal models from the aspects of model animal selection, rheumatoid arthritis model types, modeling drugs, and detection indicators, so as to promote in-depth research on rheumatoid arthritis.
