Objective To explore the method of intraperitoneal injection of different doses of cisplatin to establish a rat model of chemotherapy-injured premature ovarian failure.
Methods 48 female nulliparous Wisar rats were divided into 6 groups with 8 rats in each group. Different doses of CP were injected intraperitoneally at different time points. Control group (group A) without treatment; CP intraperitoneal small-dose (1.5 mg/kg) injection group 1 (group B), low-dose (2.0 mg/kg) injection group 2 (C group) and low-dose (2.5 mg/kg) injection 3 groups (group D), all once a day for 5 consecutive days; CP was injected intraperitoneally with high-dose group 1 (group E), 4 mg/kg, and injected again at 1-week intervals with 4 mg/kg; CP was injected intraperitoneally with high-dose group 2 (group F), 4 mg/kg, 6 mg/kg at one-week intervals to establish a chemotherapy-injured POF model in rats. The general condition and estrous cycle of the rats were observed, the changes in body weight and serum anti-Müllerian hormone (AMH) levels, as well as the pathological changes of the ovary and the number of follicles at all levels were observed.
Results Compared with the group A, the estrous cycle of the rats in each administration group was obviously disordered, the body weight was significantly decreased, and the serum AMH level was also significantly decreased (P<0.05). The number of follicles at all levels decreased significantly (P<0.05), and the number of follicles at all levels in group E and group F decreased more significantly (P-0.01).
Conclusions Intraperitoneal injection of 4 mg/kg CP in rats, followed by re-injection at intervals of one week, can induce better modeling effect of chemotherapy-injured POF, and the modeling method is simple.
