【Animal modeling】-Establishment of long-term depression mouse model and observation of behavioral and neuropathological characteristics

Objective To construct a new long-term depression model mouse, and compare the advantages and disadvantages of this mouse model and the common depression mouse model.

Methods The LTD mice were constructed by early mother-infant separation and persistent chronic stress after weaning, and compared with chronic unpredictable wild stress (CUMS) and early life stress model (ELS) ) mice with long-term depression-like behavioral changes, and compared the changes in hippocampal neurons in eGFP transgenic LTD mice and control mice.

Results Compared with CUMS and ELS mice, LTD mice exhibited a more stable depressive state behaviorally for a longer period of time. Histological sections showed that the hippocampal CA1 and CA3 regions of LTD mice were degenerated.

Conclusion The LTD mouse was successfully constructed. This model mouse can lead to longer-term depression-like behaviors and show a long-term psychological stress state. It is a model mouse suitable for long-term simulation of depression.
