Objective: To detect the effect of low concentration of levofloxacin on the pneumonia model of Pseudomonas aeruginosa (PA)-infected mice, and to explore the method of levofloxacin drug resistance.
Methods: The PA-infected mice model of pneumonia was used. In each experiment, mice were randomly divided into groups according to their body weight. Different bacterial concentration, infection times and administration methods were used. The treatment effect of levofloxacin and the feasibility of model establishment were discussed. Results The lung index of model group 1 and model group 2 were significantly different from those of the normal group when low concentration levofloxacin had a single infection of PA (1×109cfu/mL) in mice ( P<0.01); there was no significant difference between levofloxacin group 2 and model group 2. Low concentration of levofloxacin to PA (1×1010cfu/mL) single-infected mice, model group 1 and model group 2 compared with the normal group, both There was a significant difference (P<0.01). There was no significant difference between the levofloxacin group 3 and the model group 2. The levofloxacin groups 1 and 2 could significantly reduce the lung index of the model group; in the PA repeated infection mouse experiment, the model group 1 Compared with the model group 2 and the normal group, there were significant differences (P<0.01). There was no significant difference between the levofloxacin group 2 and the model group 2. In each experiment, the weight of the mice on the second day after infection was higher than that of the normal group decreased, and the results were not significantly different.
Conclusion: Under the experimental conditions, low concentration levofloxacin to PA (1×1010cfu/mL) single-infected mice, low concentration levofloxacin to PA repeated infection in mice, the animal model can simulate the phenomenon of drug resistance in clinical to a certain extent.
